can the IPv6 reverse DNS record be updated via the API? I am seeking a REST type of IPv6 reverse DNS record update solution.

The API does allow you to create and update DNS records, including IPv6. Though, we do not support setting rDNS directly for either IPv4 or IPv6. rDNS (PTR ) records are automatically configured for your IPs based on the hostname that you provide for your droplet. For IPv6, it is only for the first address that is assigned.
Is it possible to update more than just the first IPv6 PTR? It’s good to have a reverse DNS for the first address, but the other 15 don’t have a PTR.
