I wondered how you can use DO spaces as the main storage for nextcloud. Do I have to configure Nextcloud’s configration to run all data on an Amazon S3?

Yes! Spaces were designed to be compatible with S3 in order to take advantage of existing integration just like this.

DO:Spaces is S3 compatible so you should be able to accomplish this.
