Is it possible to connect the DO managed Database in plesk as default Database Server?

If you’re using the one-click app from the marketplace this will not be an option. The one click Plesk install comes with Maria DB 5.5, 10.0, 10.1, 10.2

What I can think is to do a manual installation of Plesk in order to do that. May I ask what is your main idea (ultimate goal) about using the managed database product? We might be able to see which is the best possible solution for your needs.
